Intramolecular glycosylation
Carbohydrate oligomers remain challenging targets for chemists due to the requirement for elaborate protecting and leaving group manipulations, functionalization, tedious purification, and sophisticated characterization. Achieving high stereocontrol in glycosylation reactions is arguably the major hurdle that chemists experience. This review article overviews methods for intramolecular glycosyl...
متن کاملImaging Cell Surface Glycosylation in Vivo Using “Double Click” Chemistry
Dynamic alterations in cell surface glycosylation occur in numerous biological processes that involve cell-cell communication and cell migration. We report here imaging of cell surface glycosylation in live mice using double click chemistry. Cell surface glycans were metabolically labeled using peracetylated azido-labeled N-acetylgalactosamine and then reacted, in the first click reaction, with...
متن کاملMonitoring Dynamic Glycosylation in Vivo Using Supersensitive Click Chemistry
To monitor the kinetics of biological processes that take place within the minute time scale, simple and fast analytical methods are required. In this article, we present our discovery of an azide with an internal Cu(I)-chelating motif that enabled the development of the fastest protocol for Cu(I)-catalyzed azide-alkyne cycloaddition (CuAAC) to date, and its application toward following the dyn...
متن کاملSurface glycosylation of polymer membrane by thiol-yne click chemistry for affinity adsorption of lectin.
We present a novel approach to constructing glycosylated surface for microporous membrane. Carbohydrate derivative can be facilely bound onto the alkyne-modified membrane surface via thiol-yne click chemistry. The glycosylated membrane surface shows an excellent affinity adsorption to lectin on the basis of carbohydrate-protein recognition.
متن کاملClick chemistry for the synthesis of RGD-containing integrin ligands.
In the last few years click chemistry reactions, and in particular copper-catalyzed cycloadditions, have been used intensively for the preparation of new bioconjugate molecules and materials applicable to biomedical and pharmaceutical areas. This review will be focused on conjugates of the tripeptide Arg-Gly-Asp formed by means of click chemistry reactions. This sequence is a well known binding...
